Product Description

Thioglycolate (thioglycolic acid) is used in water testing as a powerful reducing and complexing agent to measure both dissolved and total iron. It works by reducing insoluble ferric iron (Fe³⁺) to ferrous iron (Fe²⁺) and binding with it to form a highly visible purple complex.

Thioglycolic acid is used for the reduction of iron(III) to iron(II), and for pH adjustment. Thus only one reagent is required for analysis.

How the Thioglycolate Method Works

 

  • Reduction: Thioglycolic acid reduces oxidized ferric iron (Fe³⁺) into soluble ferrous iron (Fe²⁺).
  •  

  • Complexation: The ferrous iron reacts with the thioglycolate to form a purple or pink-colored ferrous thioglycolate complex.
  •  

  • Measurement: The intensity of the resulting color is directly proportional to the total iron concentration in the water sample
  • Typical Testing Procedure
    1. Color Comparison: After a brief waiting period (usually 3 to 5 minutes), the color of the sample is compared against a standard color chart or measured photometrically to determine the iron level in mg/L (or ppm).

  • Sample Preparation: A measured water sample is placed into a test tube or flask.
  •  

  • Reagent Addition: Citric acid is often added first to prevent the precipitation of iron, followed by the thioglycolate indicator.
  •  

  • pH Adjustment: Ammonia is added to make the solution alkaline. This step is crucial for the colored complex to form.

    A thioglycollate indicator (typically resazurin) is a redox dye used to visually map oxygen levels in bacterial culture tubes. 
    In its oxidized state, it turns pink when exposed to oxygen; in its reduced, oxygen-free state (typically at the bottom), it remains colorless.
     
    Resazurin Indicator Characteristics
    Pink Zone: Indicates the presence of oxygen (aerobic zone) at the top of the tube.
    Colorless Zone: Indicates the absence of oxygen (anaerobic zone) deeper in the tube.
    Reversibility: Resazurin is highly sensitive; if the broth is shaken, it re-oxidizes and the top portion will turn pink until the thioglycolic acid reduces the oxygen again

    2,4,6- Tris-(2’ - Pyridyl )-s-Triazine ( TPTZ ) or  also can be use like 2,4,6-Tripyridyl-s-triazine. Solution of (TPTZ), .001 M were prepared by dissolving 0.312 gram of compound in a view drops of hydrochloric acid and diluting to 1 ltr with water.

     
    2,4,6-Tripyridyl-s-triazine solution of (TPTZ) is useful for the calorimetric determination of iron. The Intense violet colour formed by TPTZ with iron(II) is due to the ion {Fe(TPTZ)2}+2, with only two of the three pyridyl group of each TPTZ ligand attached to the Iron atom.
     
    This reagent is  also used for determination of iron in silicates, burnt refractories, and limestone etc
